AD:The Laboratory Medicine Center ,Affiliated Hospital of Nantong University , Nantong 226001 , China Corresponding author :WANG Hui2min , E2mail : ntfyjyk @pub. nt . jsinfo. net 【Abstract】 Objective To investigate the gene mutation in the areas of pre corePcore (Pre CPC) and basic core promotor (BCP) of HBV DNA and its clinical significance1 Methods The nt 1 73521 965 segment of HBV DNA was amplified with PCR in 54 cases with chronic hepatitis B and 10 cases with post2hepatitis cirrhosis1 Then the PCR product was sequenced1 Results There were 168 site mutations in 4815 % (33P68) cases with hepatitis B1 The first ten mutation sites were nt 1 764 (5818 %) , 1 762 (4815 %) , 1 799 (2110 %) , 1 766 (1417 %) ,1 896 (1312 %) , 1 754 (818 %) , 1 899 (818 %) , 1 768 (714 %) , 1 814 (714 %) and 1 913 (714 %) 1 Three rare mutations of nt 1907 ,1 922 and 1 923 were also detected1 The mutations of nt 1 896 , 1 764 and 1 762 were found in 1617 % , 3512 % and 3512 % of chronic hepatitis , and in 3010 % ,6010 % and 6010 % respectively of post2hepatitis cirrhosis cases1 There was statistical significance between the two groups ( P < 0101) 1 Conclusion The mutations in the areas of Pre CPC and BCP of HBV DNA might possibly be associated with liver fibrosis1 There are many mutation sites in HBV DNA and mutation occurs frequently , therefore gene sequencing is helpful to the design of gene chip and to clinical application1 【Key words】 DNA; Hepatitis B virus ; Genes ; Polymorphism ,restriction fragment length ; Mutation
