TI:Assessment of allergenicity of genetically modified rice S86 by BN rat model

Abstract : To assess the allergenicity of the genetically modified (GM) rice S86 , 4 groups of BN rats (10 for each group) were fed with different experimental diets for six weeks. A positive control group and a negative control group were fed with standard AIN - 93Gdiet ; the GM group was fed with AIN - 93G- based diet containing GM rice 7813 %; the non2GM group was fed with AIN - 93G - based diet containing non2GM rice 7417 %. On day 0 and day 7 , rats in the positive control group were given Ovalbumin (OVA) 100μg (1ml) intraperitoneally and rats in the other 3 groups were given normal saline of the same volume. On day 7 , im mediately after the intraperitoneal injection , blood samples were obtained for determination of the plasma level of histamine. On days 14 , 21 , 28 , 35 , 42 , blood samples were drawn and analyzed for the serum levels of lgG and lgE. Blood pressure of the rats was measured before and after the experimental period. The results showed that no lgGor lgE response was elicited in rats of the GM rice group , while strong lgG and lgE re2 sponses were observed in rats of the positive control group ; and the rats of the positive control group had sig2 nificantly higher plasma level of histamine than the other 3 groups with similar histamine level . An oral challenge with OVA caused a temporary drop of the systolic pressure in 2 rats of the positive control group , and no change in blood pressure was observed in rats fed with GM rice. It is concluded that GM rice S86 has no allergenic effect on BN rats.
